Does the valve have to pass through the full thickness of the panel?

That is how this type of unit works: a channel connects the inside of the chamber with the surroundings. The manufacturer does not give the panel thickness here, so it needs to be measured before ordering.

How often should the valve's passage be checked for clearance?

The datasheet does not state an inspection interval. In practice it gets checked at every chamber service, since a fault often only shows up as resistance when opening the door.
